Development of Green Technological Platforms Employing Supramolecular Carriers for Poorly Soluble Drugs and Nutraceuticals

| Summary | Water solubility is a key factor in determining bioavailability and the technological feasibility of pharmaceutical dosage form development. Given that as many as 90% of new chemical entities exhibit limited solubility, the development of such drugs is expected to become increasingly technologically demanding. At the same time, the limited solubility of nutraceuticals is problematic for achieving efficient extraction and adequate bioavailability and is often accompanied by their pronounced chemical instability. Various technological approaches are employed to improve the solubility of drugs and nutraceuticals, and these must incorporate sustainability and green chemistry principles to ensure regulatory compliance and reduce environmental impact. In this context, the main objective of the proposed project is to develop innovative technological platforms with high translational potential for delivering poorly soluble drugs and nutraceuticals by designing advanced pharmaceutical dosage forms based on sustainability principles and applying green technologies. The key technological strategy that will be the focus of the project activities is the incorporation of active compounds into biocompatible supramolecular carriers of polysaccharide (cyclodextrins) and lipid (phospholipid nanoparticles) origin, achieved through the integration of various formulation strategies and technologies, while applying green chemistry principles. This includes rationalising research using in silico design, using biocompatible and biodegradable excipients and green solvents, monitoring the environmental footprint of the developed processes, and focusing on secondary raw materials. Project activities will be carried out through three work packages (WPs): WP1. Integrated application of cyclodextrins and green technologies in developing orodispersible pharmaceutical dosage forms for poorly soluble anti-inflammatory drugs WP2. Sustainable approach to developing advanced topical formulations based on supramolecular phospholipid (nano)systems WP3. Development of green extraction and formulation of multicomponent nutraceuticals based on flavonoids/carotenoids using supramolecular carriers Specifically, the project results will contribute to promoting innovation, strengthening scientific collaboration, and facilitating knowledge transfer. In a broader sense, the project outcomes will make a significant contribution to the Sustainable Development Goals and the green transition, while fostering collaboration with industry. |
